Medetomidine HClcleptor is a selective alpha 2-adrenoceptor (alpha2-adrenaline receptor) agonist with a Ki of 1.08 nM, 1620 times higher than the selective action of alpha 1-adrenoceptor.
In anaesthetic rats, medetomidine (1-100 micrograms/kg, intravenously) causes a brief decrease in dose dependence on blood pressure and heart rate. In rats with cerebrospinal cord destruction, medetomidine showed effective vascular pressure (PD501.7 micrograms/kg) and cross-sensing inhibition (ID501.6 micrograms/kg) without affecting the underlying heart rate. Medetomidine causes dose-dependent sedation, causes loss of positive reflexes at high doses (>100 micrograms/kg) and lowers body temperature. Medetomidine causes a decrease in bioamine turnover in the brain, dose dependence inhibits norepinephrine (NE) flipping, inhibits dopamine flipping in the brain at high doses, and reduces serotonin flipping by 5-serotonin.
